주기적 흡착방법에 의한 과당과 포도당의 분리
Separation of Fructose and Glucose via Cycling Zone Adsorption
Eluent를 사용하지 않고 온도를 이용한 주기적 흡착방법을 이용하여, 물리화학적 유사성 때문에 분리가 어려운 것으로 알려진 과당과 포도당의 분리를 시도하였다. 포도당에 대한 이온교환수지의 흡착능은 온도의 영향을 별로 받지 않는 반면 과당의 경우 많은 영향을 받는 특성을 이용하여 이온교환칼럼을 5℃와 50℃로 교대로 가열하면서 과당의 분리능을 살펴보았다. 분리능은 온도변화의 주기가 원료용액의 유속의 함수로 나타낼 수 있었으며 온도 변화의 주기가 원료용액의 관내 체류시간의 2배가 될 때 분리능은 최고치를 보였다. 원료용액의 농도를 1.5%에서 5%로 증가시킨 결과 분리인자가 1.15에서 1.0으로 감소하였다. 칼럼내의 온도구배를 줄이기 위해 직경이 작은 여러 개의 칼럼을 사용한 결과 분리능의 향상을 볼 수 있었다.
The separation of glucose and fructose, which is very difficult because of their physico-chemical similarity, was attempted using a temperature cycling zone adsorption that does not need eluent in ion exchange chromatography. The separation of fructose was studied by heating ion exchange column to 5℃ and 50℃ alternately, using the fact that the adsorptive capacity of glucose to an ion exchange resin does not depend much on temperature while that of fructose is more sensitive to temperature. The separability was represented by the function of the period of temperature change and the flow rate of feed solution and it was maximal when the period of temperature change was twice as nuch as the residence time of the feed solution in the column. The separation factor decreased from 1.15 to 1.0 with the increase of feeding solution concentration from 1.5% to 5%. In order to reduce the temperature gradient within the column a bundle of small multicolumns were adopted to show the increase in the separability.
