Abstract

본 연구는 병풀 (Centella asiatica)에서 유래한 식물화학물질 (phytochemicals)인 asiaticoside와 madecassoside의 추출 효율에 미치는 deep eutectic solvent (DES) 조성을 최적화하였다. Ethylene glycol (EG)과 choline chloride (ChCl)를 다양한 몰비로 조합한 DES에 물을 0 ~ 30%까지 첨가하여 추출하였으며, 그 추출물을 HPLC로 정량 분석하였다. 그 결과, EG:ChCl = 3:1, 물 20% 조건에서 두 화합물의 총 추출량이 21.5 ng/mg으로 가장 높았다. Asiaticoside는 상대적으로 조건 변화에 안정적이었으며, madecassoside는 특정 조건에서 민감한 반응을 보였다. 이러한 차이는 당 구성에 따른 친수성 차이에서 기인한 것으로 판단된다. 본 연구는 DES의 물 조성 및 HBD:HBA 비율 조절이 식물 유래 극성 화합물 추출의 주요 변수임을 입증하였으며, 친환경 천연물 추출 공정 설계에 기초자료로 활용될 수 있다.
This study optimized the composition of deep eutectic solvents (DES) for the efficient extraction of the phytochemicals asiaticoside and madecassoside from Centella asiatica. DESs were prepared using ethylene glycol (EG) and choline chloride (ChCl) at varying molar ratios, with 0 ~ 30% water added. Extraction yields were quantified using HPLC system. The optimal condition (EG:ChCl = 3:1, 20% water) yielded a total of 21.5 ng/mg. Asiaticoside showed stable extraction across conditions, while madecassoside was more sensitive. These differences were attributed to the hydrophilicity of their sugar moieties. This work demonstrates that adjusting water content and HBD:HBA ratio is key to efficient DES-based extraction and offers a foundation for green extraction process development.
