특성화된 poly(hydroxyethyl-methacrylate) 비드를 이용한 human immunoglobulin G의 흡착분리에 관한 연구(Ⅱ) : Divinylsulfone-poly(hydroxyethyl-methacrylate) 비드의 human immunoglobulin G의 흡착에 대한 염과 pH의 영향
A Study on the Adsorptive Separation of Human Immunoglobulin G with the Characterized poly(hydroxyethyl-methacrylate) Beads(II): Effect of Salts and pH on the Adsorption of Human Immunoglobulin G by Divinylsulfone-poly(hydroxyethyl-methacrylate) Beads
Divinylsulfone(DVS)을 특성기로 갖는 poly(hydroxyethyl-methacrylate, HEMA) 비드의 human immunoglobulin G(HIgG) 흡착에 대한 sodium sulfate, ammonium sulfate, potassium sulfate의 sulfate 계열 염들과 sodium chloride의 농도와 완충액 pH의 영향을 고찰하였다. Sulfate 계열의 염들은 완충액에 첨가되는 농도에 비례하여 DVS-polyHEMA 비드의 HIgG 흡착을 증가시켰으나, sodium chloride 농도 변화의 영향은 없었다. 동일한 sulfate 염에서도 양이온의 종류에 따라 HigG 흡착량이 달라졌으며, 0.5 M 농도에서 Na(+), K(+), NH4(+) 순으로 흡착을 촉진시켰다. 완충액의 pH 변화는 DVS-polyHEMA 비드의 HIgG 흡착에 영향을 주었으나, 염에 따른 경향성은 일정하지 않았다.
It was were estimated how the buffer pH, the salt concentration and the salt species such as sodium sulfate, ammonium sulfate, potassium sulfate and sodium chloride affected on the human immunoglobulin G(HIgG) adsorption by using poly(hydroxyethyl-methacrylate, HEMA) beads reacted with divinylsulfone(DVS). Salts of sulfate series promoted the HIgG adsorption according to their concentrations but sodium chloride did not. In the sulfate salts, the adsorption tendencies were different with cation species and were increased in the order of Na(+), K(+), (NH4)(+). There was the effect of pH on the adsorption of HIgG, but a constant tendency with salts was not observed.
